the word FIFO in the line
FIFO: open: No such device or address
is probably the file name. I don't think it has a problem with /dev/ ttyS0.
man fifo ->
[...]
A process can open a FIFO in non-blocking mode.
[...]
In this case,
[...]
opening for write only will fail with ENXIO (no such device or
address) unless the other end has already been opened.


Files FIFO and FIFO.ttyS0 (in /var/spool/hylafax) are used for communication between Hylafax processes. According to the error message no process has opened FIFO for reading.

Do you get Hylafax messages in syslog?
Maybe you should increase the value ServerTracing in Hylafax' config to get more messages. (For debugging I canged this from the default value
0x08501 to 0x03FFFF on my system.)
